Item The Biochemistry of Trypanosomes(1960) Knight, R.H.; East African Trypanosomiasis Research OrginizationIn spite of the considerable amount of work which has been directed towards an understanding of the biochemistry of the genus Trypanosoma there are still many gaps in our knowledge. It is the object of this note to point out a few of the subjects which might repay investigation. A study of the carbohydrate metabolism of many species of trypanosome has shown considerable differences between the species belonging to different groups. For example, it has been shown that the members of the T. brucei group degrade glucose to a mixture of pyruvic acid and glycerol, the R.Q. being small, while the members of the T. lewisi group convert glucose to a mixture of lactic, acetic and succinic acids. The R.Q. in these cases is about unity. Whereas the respiration of T. cruzi and T. lewisi is very sensitive to cyanide, that of T. vivax and T. rhodesiense is insensitive. This difference in sensitivity has been correlated with the absence or presence of a cytochrome system, such a system being present in the insect and blood forms of the species which are sensitive to cyanide and absent from the blood stream forms of the members of the T. brucei group. However, examination of the insect form of T. gambiense has shown the presence of a cytochrome system.Item The Preservation of Agar Gel Electropherograms(1964) Humphreys, K.C.; Knight, R.H.; East African Trypanosomiasis Research Organization, Tororo, Uganda.The use of microscope slides as supports for the medIUm during electrophoresis on agar gels makes the preservation of the gel, by drying it on the slide, a simple matter. The following method has been found suitable for preserving larger preparations of agar on which electrophoretically separated proteins have been stained; in the work during which the method was developed the gels measured 90 x 50 x 2·5 mm. and contained 1 % agar.Item The Prevalence of the Erythrocyte Glucose -6 Phosphate Dehydrogenase Deficiency Among Africans in Uganda(1963) Robertson, D.H.H.; Knight, R.H.; East African Trypanosomiasis Research Organization, Tororo, Uganda1) A rapid screening test has been used to determine the prevalence of the erythrocyte glucose-6-phosphate dehydrogenase deficiency in East African males living in Uganda. 2) Of 498 males examined, 81 showed the deficiency. 3) Several males showed a deficiency much greater than the rest. 4) In an examination of 164 females no deficient subjects were detected by this screening test, although the group examined might have been expected to contain at least 44 heterozygotes and 4 homozygotes. 5) The importance of the variety of the dye used in the test is stressed.
